5.2.1 Remove the lid and place it carefully on the bench with the sachet uppermost, to reduce the risk of its contamination.
5.2.2 Load the cultures into the jar in their dishes or containers or tubes.
5.2.3 Keep one anaerobic gas pack of 3.5 liter pack with anaero indicator tablet and immediately replace the lid on the jar and secure it with bridge clamp, finger tightening the Bakelite knob.
5.2.4 Incubate the jar at required temperature for the required period.
5.2.5 After incubation, remove the bridge clamp and open one of the needle valves so that any reduce pressure inside the valve will be relived and the other cover may be removed and the end of the activity clean the Jar with disinfectant.
