1.0 PURPOSE:
To define the procedures for performing lacto phenol cotton blue staining technique for identification of fungi.
2.0 SCOPE:
This SOP for lacto phenol cotton blue staining for identification of fungi is applicable for the identification of the fungal colonies in quality control department at (Company name).

5.0 FREQUENCY:

6.0 PROCEDURE:
6.1 Place two-three drops of lacto phenol cotton blue stain on the center of clean glass slide.
6.2 Take a small tuft of fungal culture from the plate along with the spores with the help of flamed and cooled needle and place it on the lacto phenol cotton blue stain.
6.3 Separate the tufts into finger size using two flamed and cooled needles.
6.4 Place a neat and clean cover slip on the culture and ensure that no bubble is formed.
6.5 Observed the slide under the microscope, first under 10X lens and then under 40X lens.
